Exo Therapeutics is a stealth-mode, venture capital-backed company from Harvard University & the Broad Institute with headquarters in bustling Kendall Sq., Cambridge. Exo is developing an innovative small-molecule discovery platform that targets Exosites – distal binding pockets at the junction of enzymes. Targeting enzyme junctions in this way will reprogram enzymatic activities in sophisticated and specific ways and inhibit only the therapeutically relevant activity of the enzyme. Exo is using the technology to development small molecule drugs with a focus in oncology.
Position Summary
Exo Therapeutics seeks a highly motivated biologist with a strong background in cell-based assay development. This position will be an integral part of the biology group and will help drive the drug discovery programs forward. You will work across therapeutic functions with a primary focus in oncology to help discover and develop innovative therapeutics.
This position is predominantly laboratory/wet bench based and requires strong experimental technique, the ability to deliver quality data in a timely manner from high throughput cell-based assays and work effectively with team members in support of small molecule drug discovery efforts. Data analysis, interpretation and assay development experience is required.
Responsibilities
Key responsibilities of this role include, but are not limited to:
- Design and execute experiments, and perform data analysis
- Design, QC, validate, and execute medium- to high- throughput 96/384 well assay formats, such as ELISA, quantitative Western, high content IF cell imaging, cell viability/death utilizing small molecules to generate full dose response curves
- Leverage multiple assay platforms to measure target engagement, pathway modulation and cell sensitivity in response to compounds or target modulation
- Perform molecular techniques including DNA/RNA isolation, cloning, PCR, qRT-PCR, gene expression analysis, cell line engineering as needed
- Identify and characterize appropriate cell models that are dependent on the target of interest
- Utilize compounds and molecular target manipulation (e.g. RNAi) to dissect target function
- Interact and collaborate with biology and chemistry team members to contribute to drug discovery and development program progression, present data at regular team meetings
- Document, compile and interpret experimental data; manage laboratory notebook, databases, and/or spreadsheets to present data in a meaningful manner
Required Qualifications
- BS/MS with at least 5+ years of experience in in Vitro Pharmacology, Cell biology or a related discipline degree with significant proven experience, preferably in the Pharmaceutical or Biotechnology industry
- Prior experience working and managing CRO partnerships preferred
Experienced in-
- Aseptic tissue culture techniques and mammalian cell tissue culture
- Protein lysate preparation from cells and tissues, and quantitative Western blotting
- Molecular techniques including DNA/RNA isolation, cloning, and PCR/ qRT-PCR
- Cellular functional assays such as cytotoxicity, apoptosis, and siRNA/shRNA transfection
- Working with small molecules to generate full dose response curves in drug discovery
- Development and optimization of high throughput (96/384 well) cell-based assays using a variety of readout technologies (e.g. fluorescence, luminescence, AlphaLISA)
- Various bioassays such as High content IF imaging, ELISA, multiplex protein analysis is preferred
- Ability to work as a team player and be able to work on several projects simultaneously
- Understanding of oncology and/or immunology is desirable

Overview
Exo Therapeutics seeks a highly motivated senior/principal research associate with a strong background in biochemistry assay implementation and contributing to a multidisciplinary team in external and internal drug discovery activities. This position will be an integral part of the biochemistry team and will help drive the drug discovery programs forward. You will work across therapeutic functions with a primary focus in discovery and development of therapeutics in oncology, autoimmune and immuno-inflammation.
The senior/principal research associate will be expected to contribute to and complement our existing technical expertise to enhance and accelerate research. We are keen to identify someone who is a self-starter and proactive and who can work independently and effectively, both with the Exo team and external collaborators. The candidate will also participate in the build-out and maintenance of our growing laboratory environment.
Responsibilities
- Perform routine biochemical screening of compounds, eg IC50, in house or in collaboration with external partners.
- Communication and discussion of results with internal project teams, especially chemistry and biology.
- Development of novel screening assays for use either in-house or for transfer to CRO’s.
- Coordination of assay workflow locally and/or with the CRO partners, data analysis and presentation of results to the internal and external teams.
- Employ biochemical and biophysical methods enabling mechanistic studies to determine the mode-of-inhibition for promising hits.
- Propose and implement new experimental techniques and troubleshoot roadblocks in collaboration with the team.
- Independently manage and order supplies/reagents in a timely manner to ensure continuous progress of parallel experimental workflows under your responsibility.
- Management of laboratory equipment, including maintenance, vendor relations and service contracts.
- Maintain good data records, protocols, and summaries. Analyze raw data and interpret integrated datasets using Prism, Vortex, or similar software packages.
- Use of electronic lab notebook and data registration to corporate database.
- Take all opportunities to make intellectual contributions and discoveries.
Required Qualifications
- Bachelor’s degree in a life sciences-related field with by a proven track-record of hands-on laboratory experience for 4-6 years, or a Master’s degree with 4+ years of research experience, respectively.
- Strong foundation with a background in small molecule discovery/validation, biochemistry and biophysical methods.
- Proficiency in experimental design and data analysis using Graphpad Prism, Excel or equivalent software.
- Troubleshooting skills, analytical perseverance, with a strong ability to integrate advice.
- Proficiency in results presentation, data documentation, and scientific communication.
- Analyze, interpret, and effectively present scientific data at team meetings
- Ability to work effectively with flexible supervision as a member of multiple project teams
- Prior experience working with CRO partners highly preferred
- Good problem-solving skills with excellent interpersonal skills and a collaborative work style
Desired technical competencies:
- Assay development experience and capabilities such as FP, TR-FRET, AlphaScreen, etc.
- Experience in mechanistic enzymology such as Ki, mode-of-inhibition, kinetics, etc.
Other complementary and transferable skills:
- Molecular biology, protein expression, purification and characterization (for example: M/S, SEC, DLS, CD, SDS-PAGE, etc.
- Working knowledge of biophysical methods such as SPR, MST, ITC, DSF, etc.
- Familiarity with Cell-based assays (for example: BRET, iLite, DualGlo, CTGlo, HiBiT, CaspaseGlo, etc.)
